MacPherson Suspension Kinematics Analysis

This bachelor thesis deals with the influence of the position of significant points of MacPherson strut on kinematic characteristics. The first part contains a description of selected suspension parameters, their impact on the driving properties of the car, and a description of MacPherson strut. Furthermore, based on provided data, is prepared a detailed analysis of the influence of kinematics points to individual characteristics. In conclusion, all findings are synoptically summarized so that they can serve as a manual for change of the kinematic characteristics.

Proposes two buildings located at the top of the raised part of the resolved region. One building is positioned parallel to the access road. The shape of the building recalls the long kubus facing in the direction of the driveway and the building of accommodation is designed perpendicular to the object. Both objects are embedded into the basement terrain. Accommodation building is situated on the south, bright and warm side with a beautiful view.
Engineering design of a fullsuspension mountain bike frame

The aim of this bachelor thesis is designing of a full-suspension mountain bike frame. At the beginning of the thesis mountain bikes are divided into categories, the function of a shock absorber is described and issues of bike kinematics and related characteristics are clarified. The thesis also describes properties of the most used materials for bike frames manufacturing. Subsequently, the most common systems of rear wheel linkage are analysed. Next, the requirements for own design are identified in terms of geometry, construction, kinematics and a creation of its 3D model in SolidWorks software. The characteristics of designed frame are analysed by specific software Linkage X3. Then a static analysis in Ansys software is made in order to select bearings for individual pivots. The final part of the thesis is dedicated to visualisation of designed frame.
Návrh tematické turistické trasy v mikroregionu Kahan

This bachelor thesis deals with a suggestion of a theme-based walking trail for tourists. The goal of this thesis is to design a trail for a tourist educational nature trail in the Kahan microregion, based in the topic of a novel Pohádka máje, to design supporting materials and estimate the price of construction of the walking trail. The trail and its target group were designed in cooperation with the Kahan microregion. The preconditions of local tourism in the Kahan microregion were stated, the existing educational nature trails of the Kahan microregion were mapped and SWOT analysis of the suggestion was carried out. The result is a suggestion of the trail „Travels to Vilém and Helenka“ as well as information leaflet drafts and a worksheet. The conclusion offers possible options for future redesign of the trail and includes the estimated price of the construction of the route.
Apoptosis of tumor cells : role of TRAIL and caspase 10

One of the key features of cancer cells is the ability to escape programmed cell death (apoptosis). As a mechanism of apoptosis inactivation in cancer cells, somatic mutations of pro-apoptotic genes have been reported in many cancers. Caspase 10 is an initiator caspase whose physiological function remains poorly understood. Also the ability of caspase 10 to substitute for caspase 8 in the death receptors apoptotic pathway is still controversial. However, the fact that some of the mutations found in CASP10 gene was associated with apoptosis defects (79, 81) suggest that caspase 10 could be also important in apoptosis initiation. In our lab, there was found a heterozygous mutation in CASP10 gene of Jurkat (human T-acute lymphoblastic leukemia) clone resistant to TRAIL (J-TR1). This mutation influence the amino acid composition close to the active site of the enzyme. The aim of this thesis was to confirm the mutation by ARMS-PCR and to determine if an overexpression of normal (unmutated) or mutated caspase 10 D in TRAIL sensitive and/or TRAIL resistant Jurkat cells (J-WT and/or J-TR1) will influence TRAIL induced apoptosis. The thesis contains a table as a specific space, which connotes different meanings in the context of contemporary events. The theoretical part refers thus to the social relations, which are part of both public and private - family space in which a table plays its essential role. Not only as a subject form of everyday life, which is linked to a personal experience, but also as a form of symbolic expression. A table is kind of bearer of communication, message. This will be supported not only via etymological and sociological analysis, but also by the analysis of selected works of art. It is a way to inspect this phenomenon in the aforementioned context. The aim of this study is to show - through fine arts, historical and sociological relations - the view of a table as a continuously interconnected complex, through which it is possible to better realize some family ties and social events, including contemporary needs and priorities.. Canonical and non-canonical signalling triggered by activated TRAIL receptors in human cells

TRAIL ligand can trigger apoptosis of permissive human cells via engagement of its two pro- apoptotic receptors TRAIL-R1 (DR4) and TRAIL-R2 (DR5). Its ability to induce apoptosis independently on p53 status and to selectively kill cancer cells in vitro and in vivo made this ligand an attractive target in cancer research. However, acquired resistance of primary cancer cells, unsatisfactory outcome of clinical trials and recent studies arguing that TRAIL might under specific conditions promote cancer progression, opened new plethora of questions, which need to be addressed. Though both receptors DR4 and DR5 are ubiquitously expressed, different types of tumours show preference for either of the receptors. The relative participation of DR4 and DR5 in TRAIL- induced signalling is still largely unknown. To analyse TRAIL receptor-specific signalling, I prepared Strep-tagged, trimerised variants of recombinant human TRAIL ligands with high affinity for either DR4 or DR5 receptor. Using these receptor-specific ligands, I examined a contribution of individual pro-apoptotic receptors to TRAIL-induced signalling pathways. Apoptosis serves as a natural barrier to cancer development, and the resistance to apoptosis represents one of the key capabilities acquired during tumor development or progression. Impairment of the intrinsic apoptotic pathway exemplifies one of the established mechanisms of constitutive or acquired drug resistance. As most of the currently used cytotoxic drugs initiate tumor cell death by direct or indirect triggering of the intrinsic apoptotic pathway, impairment of the intrinsic pathway is associated with therapy failure. Targeting of the death receptors, however, enables induction of apoptosis even in the chemotherapy resistant cancer cells. TRAIL is a death ligand belonging to the TNFα superfamily that specifically kills tumor cells while sparing healthy tissues. Much enthusiasm has been generated for TRAIL as a highly promising targeted anti-cancer agent. However, many primary tumors have been shown to be TRAIL resistant. In attempt to overcome such an intrinsic TRAIL resistance a wide array of agents have been shown to sensitize tumor cells to TRAIL. Molecular mechanisms responsible for regulation of apoptosis in prostate cancer cells treated with TRAIL and chemotherapeutic drugs

Prostate cancer is one of the most common cancer-related causes of death among men. Chemotherapy is mainly used for treatment of its later stages, accompanied by unpleasant side effects. So far, the treatment of advanced stages of prostate cancer has not been sufficient, and new more effective alternatives are needed. The application of the TRAIL cytokine, which induces apoptosis in tumor cell, but is not toxic to nonmalignant cells, seems to be a promissing approach. However, TRAIL-based therapy is often limited by the emerging cancer cell resistance. Overcoming the resistance can be achieved by combination therapy of TRAIL with effective sensitizers. Within this work, a combination of TRAIL with platinum-based chemotherapeutic drugs such as cisplatin or its novel derivative LA-12 was applied in order to facilitate the elimination of prostate cancer cells. In the experimental part of this work, using Western blot and flow cytometry analysis it was shown that TRAIL in combination with CDDP or LA-12 effectively enhanced apoptosis in three human prostate cancer cell lines. This effect was accompanied with increased activation/amount of several proapoptotic Bcl-2 family proteins, while no changes in the level of the receptors for TRAIL were observed.
